Effects of NK Cells Activity and Cell-Mediated Immunity of Ganoderma lucidum Water Extracts in Mice Transplanted with Tumor Cell(Sarcoma-180)



Abstract
These studies were attempted to assess the natural killer cells cytotoxity and cellular immunity effect of crude Ganoderma lucidum water extract(GLW) in vivo.
The crude extract was prepared by reflux extraction with distillated water for 12hrs from Ganoderma lucidum.
Every experiment was done with sarcoma-180 solid tumor in ICR mice, oral administration method. ICR mice(23¡¾2g0 were divided into two groups having 20 animals eaceh.
Then 1¡¿10E6/m§¤Sarcoma tumor cells, which had been collected from a mouse on the 15th day after implantation and cunted, were dorsal-subcutaneously inoculated into the right inguinal region of each ICR mouse with a syringe.
From 24hrs thereafter, 80mg/kg of a sample dissolved in physioiogical saline was oral administered once a day for 15 days, but control group was administered physiological saline only.
To measure NK cells cytotoxity using 4hrs 51Cr release assay, the lymphocytes were separated from the spleen of tumor-bearing mice administrated GL-W 80 mg/kg/day or ally (per os) for 15 days. And to observe antitumor effect, the mice were
sacrificed
and the solid tumors were excised from mice, average tmor weight and suppression ratio were calculated.
We used modified Boyden chamber to evaluate chemotactic activity of leukocytes from the mice of control and tested groups.
@ES The following results were obtained :
@EN 1. In the case of NK cells cytotoxity on the target cells the mice treated with GL-W(1.58¡¾0.14 UL) showed approximately three times stronger than the control group(0.46¡¾0.18 UL) (p<0.05).
2. In the case of the weight of tumor formed to the right growing of the mice injected sarcoma-180 subcutaneously and treasted with GDL-W(0.85¡¾0.30), it showed inhibition ratio of 31.45% against solid tumor cell comparing to the control
group(1.24¡¾0.29( (p<0.25).
3. The chemotactic activities of polymorphonuclear leukocytes from GL-W (83.68¡¾24.50) treated tumor-bearing mice was significantly inceased than the control group (45.90¡¾33.73)(p<0.025).
4. Differential white blood cell counts from tumor bearing mice treated with Ganoderma iucidum was no significant difference comparing to the control group. According to the above results, it indicates the Ganoderma lucidum water extracts could
inhibit
the growth of mouse tumor cells. 180) through the activation of chemotactic leukocytes and the increase of NK cell cytotoxity.
For these reasons, Ganoderma lucidum water extracts could clearly enhance to cellular immune response in mouse model experimental results.
